Leptin receptor (Leptin R), also known as B219, OB receptor (OB-R), or CD295, is a 150 kDa protein belonging to the class I cytokine receptor family. Mouse Leptin R is synthesized as a 1162 amino acid (aa) polypeptide composed of a signal peptide, an extracellular region with cytokine receptor homology (CRH), multiple fibronectin type III domains and a WSXWS motif, a transmembrane domain, and a cytoplasmic domain linked to JAK/STAT pathway. Mouse Leptin R shares approximately 78% and 93% aa sequence identity with human and rat Leptin R, respectively. Several alternative spliced forms (a-f) of obR gene have been identified. ObRa, ObRc, ObRd and ObRf with truncated cytoplasmic domains, are responsible for mediating Leptin binding and endocytosis, but not signal transduction, while ObRb (formerly OB RL) containing a large cytoplasmic domains, expressed mainly in the hypothalamic arcuate nucleus, is essential for signal transduction. Mutations of ObRb can cause obese phenotypes in both the mouse and rat. In addition, a soluble Leptin R has been identified and is the primary Leptin-binding protein in blood to maintain a pool of available bioactive Leptin, delay Leptin clearance from circulation, and down-regulate blood brain transmission of Leptin.
